The following has been kindly supplied by the Centre National de Genotypage and formed part of an application note produced in conjunction with TAP. It provides an illustration of how BasePlate is being used within the genotyping environment.
BasePlate in use for high throughput PCR set-up
PCR is a central tool for high throughput genomics analysis. As the throughput requirements in areas such as genotyping, expression studies, association studies and sequencing increase, the set-up and performance of large numbers of PCRs at low volumes becomes a bottleneck in these processes.
The photograph below illustrates the products from a high speed PCR set-up in a 5ul final volume in a384-well plates. BasePlate’s flexibility can be extended to produce PCR reactions in 32x384 well plates, allowing for the set-up of 12,000 reactions in under 1 hour.
The PCRs(1) were set-up with alternate wells containing template DNA. The resulting PCR products were run out on 3% agarose gels:

This demonstrates the consistency and success of the PCR set-ups – the BasePlate liquid handling system ensuring reliable and reproducible results with no cross contamination.
The PCRs were executed on fragments in the human B-2-adrenergic receptor gene, which belongs to a family of seven-transmembrane receptors(2). SNPs in this receptor are associated with a genetic predisposition to hypertension and are found to be relevant for cardiovascular disease(3)
