Piccolo - system overview
  There are two main aspects to Piccolo, the culture of recombinant proteins(1) and the processing of these cultures to provide a range of possible outputs from cells through to purified protein(2).
  The diagram below illustrates a typical process flow for both microbial and insect cells.
Processes automated in the Piccolo system
 
¹The culture system provides a range of culture parameters at the level of individual microbial or insect cell cultures. Variables that can be evaluated on a single run include:
  • Pre-induction and expression temperatures
  • Time of harvest
  • Inducer concentration
  • Timing of induction based on culture density
  • Media and antibiotic types
  • Multiple microbial host or insect cell lines
  • Multiple microbial expression constructs or recombinant baculovirus
²The downstream processing system in Piccolo is designed for the reproducible delivery of a range of alternative outputs from culture volumes of 10 to 60mls for E.coli and 12.5 to 75mls for insect cells.
